A lot of stuff has happened in The Witcher 3: Blood and Wine by the time you get to this quest. There have been some smaller quests as well as some major bloodsheds, there have also been some more important ones and some less important ones as well.

The Witcher 3: Blood and Wine The Night of Long Fangs Quest Walkthrough
Right after the last quest ends, this one begins with a battle with a Bruxa, which is going to be tougher to beat than the last one because it is at a higher level. However, you can continue using the same strategy you did with the last Bruxa.
Talk to Regis after this and go into the conversation knowing that you will be making an important choice here – before which, it would be a good idea to make a new save.
Both choices take you to Beauclair, and while you are going make sure to keep an eye out for vampires around you.
You will come face to face with a Gharkain that gets aggressive with lesser health. It will be at the other side of the bridge so prepare for it before that. The more you hit it, the more aggressive it will be and start using its screech and a leaping attack more.
The screech can stun you for a good long time, so you should keep safe distance unless you are really beating it into pulp. You can roll sideways to avoid attacks but not the screech.
That being said, if at any point the fight is taking its toll on you, just roll back to the bridge and away from the Gharkain.
Once you beat it, another Bruxa will come to fight you in the street; deal with it using a Yrden sign as soon as possible – for perfect timing, use it when you see a figure with a hood banish near a fire.
This is not all, a Fleder attacks you, use a combination of dodges, Igni and quick attacks on it and it won’t be much of a problem. All you need to keep in mind is that it can leap.
After this fight, you should follow the blood trail and check the bodies using Witcher senses. Be careful when you are near the barricade as soon a Bruxa and an Alp will attack. Let them take on the village people and wait for the moment when you can take on them one at a time. Use the same tactics on both.
Your fight with the Bruxa and the Alp leads to a diverging point in the game. You ending choice will also be affected by which path you choose and hence which quests you take.
If you want to ask Orianna about whereabouts of the Unseen Elder you have to go southeast, fight another Gharkain and find Orianna that will kickstart Blood Simple quest.
ALternatively, you can use your senses to follow Damien and his unit. Take the path to north, beat the bats, examine a body and kill another Fleder to start a conversation. When damien shares the information he has, talk to the ravens to start Beyond Hill and Dale.
